生物体中基因以及下游的蛋白质调控疾病的发生,以及基本的生理过程。因此通过正向遗传学技术鉴定调控疾病或者基本生理过程的基因将极大推进对于生理/病理过程的了解,并为寻找有效治疗药物提供基础。本中心将利用小鼠ENU突变技术产生大量带有基因组随机突变的小鼠,结合各实验室感兴趣的试验系统,提供并筛选出相比野生型小鼠具有不同表型的突变小鼠,并进行传代验证。对于稳定遗传的具有表型的小鼠,我们将为大家设计引物进行基因分型测定,并联系相关测序机构进行外显子组测序技术测序,分析各突变基因与表型变化的关联度,最终鉴定调控该表型变化的基因,从而为研究其功能打下基础。

Genes and downstream proteins in organisms regulate pathogenesis and basic physiological processes. Therefore, identification of genes regulating these processes via forward genetics technology will greatly enhance the understanding of the mechanism, and will pave the way for developing new therapeutics. Our center will utilize ENU mutagenesis technology in mice to generate a lot of mice with random mutations in genome, while combining with assay systems that other labs are interested in to provide and screen mutant mice with distinct phenotypes compared to wild type mice. And then inheritability tests will be performed and the phenotypes can be validated. For mutant mice that exhibit stably inherited phenotypes, we will design primers for you to conduct genotyping of these mice and will contact sequencing center to perform exome sequencing. Then we will conduct linkage analysis to assess the association between mutant genes and phenotypic changes, and finally identify the genes regulating the changes and thus lay the foundation of investigating the functions of these genes.
